Witch Pinion gear would I use for the rc10 graphite using a RS540 johnson & a 78 spur gear

Assuming your running a stealth tranny I'd start somewhere around a 20-22 for general running. There's a lot more than just what motor your running that plays into the right ratio though; running on dirt or grass, track or yard, etc

I will be running in a 4 hr enduro on a medium hardness track, I was going to try 25 pinion

Oh wow! That's cool. Well then I would lean toward the power teeth then and not get your motor too hot. Ideally on a track you want to top out about 2/3-3/4 of the way down the longest straight and the go up or down a tooth depending on what you need for the rest of the track (lower tooth if there's tighter corners or higher if it has a lot of straights and wide sweeping corners where you can keep your speed up). The most important thing to keep in mind isn't blazing fast laps it's keepng the car cool and running.
